Steps to prepare a genomic library are as follows:
  • In order to prepare a genomic library, the organism's DNA needs to be extracted from the cell.
  • Next, the DNA is digested with a restriction enzyme.
  • This ensures that the DNA is cut into fragments of the specific size. The fragments are then inserted into the vector using DNA ligase.[1
  • The fragments of DNA are collected and stored for their use in the molecular cloning.
  • This DNA is stored in vectors containing various fragments of inserted DNA.
  • These vectors are then maintained in the host cell mainly bacteria like E.coli.
  • This allows easy amplification and extraction of the clones.
